Automated High Throughput Nucleic Acid Isolation Based on Proprietary M-PVA Magnetic Bead Technology.
In cooperation with the Forschungszentrum Karlsruhe GmbH, chemagen has developed an innovative robot to realize high throughput sample preparation based on its M-PVA Magnetic Beads.
The chemagic Magnetic Separation Module I is the complete solution for preparation of nucleic acids from various sample material, such as blood, tissue, bacteria, food, plants, etc. Due to it's flexibility it can be applied e.g. for PCR-product purification, Genotyping, Virus Detection, Forensic, GMO-testing and HLA-typing.

Why use the chemagic Magnetic Separation Module I
Classical methods of DNA and RNA isolation are based on column or precipitation methods. These techniques require centrifugation or vacuum steps and often have lengthy processing times as well as volume limitations, complicating their integration into automated high throughput processes. Additionally, the automated processing of these methods may cause cross contamination of the purified nucleic acids making this method useless for many diagnostic purposes.
Another problem applying automated magnetic bead based separation technologies is the insufficient or ineffective resuspension of the magnetic bead pellets after magnetic separation to enable sufficient washing and elution steps. This ineffectiveness results in lower yields and purity of the isolated nucleic acids.
The chemagic MSM I uses a unique technology to avoid these emerging problems. In the chemagic system, magnetic separation is achieved through the use of an electromagnet and separation heads with magnetizeable rods. The metal rods are immersed into the magnetic bead suspension. When the electromagnet is switched on, the rods become magnetic and the beads are separated. To resuspend an once separated magnetic bead pellet, the magnet is switched off and a stirring motor to which all rods are connected is switched on. This leads to a very effective and gentle resuspension process resulting in separation products with both highest yields and purities (see animation).
Obstacles and Solutions at a Glance
While screening the existing technologies chemagen identified and solved the following obstacles of automated nucleic acid separation:

- Additional high costs of disposables

In most cases sample preparation is the bottleneck in HLA registry procedure. While diagnostic methods like SSO, SSP and SBT can be carried out simeltanously and cover a high sample throughput, commonly used DNA separation methods require hands on time and often don't produce reliable and constant results. The chemagic Magnetic Separation Module I bridges this gap delivering high yield und quality DNA - fast and fully traceable. |

The downstream applications utilized in human genetic studies have to rely upon quality and stability of the purified DNA. Besides, while detecting mutations often high volume samples are needed to isolate a sufficient amount of DNA for the following detection reactions. The chemagic MSM I is a perfect solution to this challenge. The quality and robustness of the purified DNA is proven in downstream applications such as SSCP (Fluorescent, Phast System), WAVE dHPLC and fragment analysis. |

The chemagic Magnetic Separation Module I can be used with exchangeable heads which allow the processing in 96- or 48-well-plates using a head with 96 magnetizable rods, in 24-well-plates using a head with 24 rods or in larger tubes (working volumes up to 50 ml) using the chemagic 12 Rod Head. The resulting flexibility makes it possible to meet individual requirements for special applications. General
The use of the chemagic MSM I simplifies and fastens time consuming sample preparation steps like nucleic acid isolation significantly. By using the chemagic Dispenser it is even faster and more comfortable.
The chemagic Dispenser was developed to notably reduce hands-on time preparing a chemagic MSM I run – especially when larger volumes are required. But independent of the amount of needed buffer this particular add-on prefills fast and accurately the tubes/plates during a chemagic MSM I run.
This precise technology of the chemagic Dispenser is also integrated in the chemagic Prepito. |

The first three points of the manual procedures can be avoided using a liquid handling robot. The empty racks are then positioned on the robot platform.

ID Window
Within the "ID" Window the primary sample ID´s can be registered manually or by scanning the barcoded primary tubes. Another or the identical code can be assigned to the corresponding elution tube. The upper part of the window shows schematically the x-axis of the instrument with an allocation of each kit componentto be used at its specific position. Identification of each Individual rack creates a further option. |

Status Protocol
The status protocol shows within the main window: the running time, remaining time and the actual protocol step while running the protocol enabling the complete tracking of the process |

Log File Window
The log file window enables the access to all log files. The files contain all the information needed for the complete documentation of the isolation process and sample tracking. The files tamper proof/right protected and can be printed out at anytime. LIMS compatibillity is guarranteed for all systems which except plain text files. |

chemagic QA Software upgrade 5.1 available
The chemagic QA Software upgrade 5.1 is now available and promises a more convenient and flexible handling of the chemagic MSM I with/without chemagic Dispenser. It is compatible with all three chemagic Rod Heads (12, 24 and 96).
