Thermo Fisher Scientific has several phases that are designed for the retention of polar compounds, or offer alternative selectivity to traditional HPLC phases.
AQUASIL C18 Capillaries
- Excellent retention and alternative selectivity for polar compounds
- Completely stable under 100% aqueous conditions
- Ideal for LC/MS applications
High efficiency KAPPA® columns are available in 0.50mm, 0.32mm, 0.18mm and the new 0.10mm ID capillaries, and have an 8th inch stainless steel sleeve around the capillary to make them very robust for day-to-day laboratory use. They also have standard steel female endfittings, and so they can be used with standard column connectors.
AQUASIL C18 capillary columns provide excellent chromatography of difficult-to-retain polar compounds due to a unique hydrophillic endcapping. This also allows the use of 100% aqueous mobile phases, without phase collapse and loss of performance. AQUASIL C18 columns can also be used with little or no additives in the mobile phase, which makes them suitable for LC/MS applications.

AQUASIL C18 Columns

AQUASIL C18 columns provide more retention for difficult-to-retain polar compounds due to a unique hydrophillic end-capping. The combination of a moderate coverage C18 group and an additional polar group gives new selectivity compared to a traditional C18. AQUASIL C18 columns can also be used with little or no additives in the mobile phase, which makes them ideal for LC/MS applications.

BETASIL Phenyl/Hexyl columns offer sharp peaks and excellent reproducibility

- Includes a Phenyl/Hexyl mixed mode phase for alternative selectivity
BETASIL Phenyl/Hexyl columns are based on 100Å pore size, fully deactivated type B silica to ensure sharp peaks and excellent reproducibility for the analysis of a wide range of compounds.
The 100Å pore size gives a high concentration of stationary phase coverage, which gives the columns the highest level of reproducibility. The mixed mode stationary phase of aromatic phenyl groups and alkyl C6 chains allows for a both dispersive and pi-pi electron interactions, that will often lead to alternative selectivity to a wholly alkyl stationary phase column.
BETASIL columns are also offered in a wide range of stationary phases, making them an ideal family to choose when a range of selectivities is required on a 100Å pore size silica.

BetaMax Acid Columns
BetaMax Acid columns with high surface area resulting in maximum retention
- High surface area area and bonded phase coverage for maximum retention, even in high percentage organic conditions
- Chemistries for acidic compounds
For alternative selectivity to traditional alkyl phases, we recommend you try the polar embedded phase BetaMax columns. The stationary phase contains urea groups embedded within the C12 chain, which extra interactions with samples to extend the capabilities of the phase.
BetaMax Acid is fully endcapped and based on a high surface area higly base deactivated 60Å pore size silica. The high surface area (540m2/g) gives the columns maximum retention, whilst the small pore size can also increase retention of small compounds that are able to fully access the pores. The high coverage C12 ligand gives the columns higher hydrophobic retention that other polar embedded phases, and compared to conventional phases (C18 or C8) it offers a completely new selectivity.
A polar functional group between alkyl ligands and the silica surface allows fast analysis without compromising the resolving power. BetaMax Acid is completely stable under 100% aqueous conditions, so it is ideal for 0 to 100% organic fast gradients or where a short column length is desired.

For alternative selectivity to traditional alkyl phases, we recommend you try BetaMax Base columns. The stationary phase contains cyano groups, which allow for added polar interactions with basic samples, and added retention for samples containing at least one amino group.
BetaMax Base is fully endcapped, with high surface area due to a 60Å pore size silica. The high surface area (540m2/g) gives the columns maximum retention, whilst the small pore size also increases retention of small compounds that are able to fully access the pores. The high coverage cyano phase gives the columns greater retention that other cyano phase columns, and compared to conventional phases (C18 or C8) it offers a completely new selectivity. The columns can be used in normal phase or reversed phase LC.

Fluophase PFP Columns
Fluophase columns for retention, selectivity for halogenated or polar compounds

For alternative selectivity to traditional aromatic phases, we recommend you try the perfluorinated phases of Fluophase columns. The stationary phase of Fluophase PFP contains a perfluorinated phenyl ring, where every hydrogen atom on the phenyl ring is substituted for a fluorine atom. This substitution gives the phase extra interactions with polar samples to extend its selectivity capabilities.
Fluophase PFP is fully endcapped, and is based on a highly base deactivated 100Å pore size silica. The pore size combined with the fluorinated phenyl phase gives the columns moderate hydrophobic retention, and due to the more polar fluorine atoms the columns have a different selectivity to traditional phenyl phases.
The Carbon-Fluorine bond has greater dipole that a Carbon-Hydrogen bond. This dipole character allows the phase to offer greater retention of polar and halogentated compounds, compared to a traditional phase. Fluophase PFP is stable under 100% aqueous conditions, so it is ideal for 0 to 100% organic fast gradients.

The columns are based on our metal free HyPURITY silica and, compared to other phases, produces a competely new selectivity. A polar functional group between alkyl ligands and the silica surface allows faster analysis without compromising the resolving power. HyPURITY ADVANCE - resulting in exceptional chromatographic performance.
The phase chemistry allows three interaction mechanismswith samples:
The polar character assists retention of polar compounds and limits interactions of samples with the silica surface - which reduces retention of basic compounds.
This effect often resukts in a completely new elution order when compared to traditional alkyl bases chemistries, and gives the columns complete stability in 100% aqueous mobile phases.
The C8 ligand gives moderate retention for neutral samples, hence HyPURITRY ADVANCE is ideal for high throughoput laboratories with complex and variable sample types.

Hypercarb Capillaries
Hypercarb capillary columns are for difficult-to-retain or resolve compounds
Due to the polar retention effect of Hypercarb, Hypercarb capillary columns provide excellent chromatography for difficult-to-retain polar compounds, or compounds that are difficult to resolve due to structural similarity.
Hypercarb capillaries are available in two popular designs:
KAPPA® and PIONEERTM direct-connection columns, which are both ideal for LC/MS.

PIONEER capillary columns minimize band spreading and improve chromatographic efficiency by connecting directly into LC/MS and other detectors with a male 10-32 connection on one end. They are available in 0.50mm and 0.32mm ID.
Due to the polar retnetion effect of Hypercarb, Hypercarb capillary columns provide excellent chromatography for difficult-to-retain polar compounds, or compounds that are difficult to resolve due to structural similarity.
Hypercarb capillaries are ideal for nano and picomol concentration LC/MS applications.

Hypercarb is a unique material for HPLC manufactured by Thermo Fisher Scientific Hypersil-Keystone. Hypercarb consists of fully porous spherical carbon particles comprised of flat sheets of hexagonally arranged carbon atoms. The carbon atoms have a fully satisfied valence and offer completely different retention and selectivity to silica and polymer based phases.
Hypercarb can be used for RP and NP chromatography with a pH range from 1 to 14, and at extremes of temperature. Hypercarb excels in the analysis of structurally related components (e.g. stereo- and diastereoisomers and positional isomers) where no separation occurs on conventional materials.
Hypercarb retains polar compounds via a 'polar retention effect' where a stronger analyte-sorbent attraction occurs as the polarity of the analyte increases, so the more polar the analyte the longer it's retention time compared to silica-based phases. Due to the unique retention mechanism of Hypercarb, we recommend that you do not use a column longer than 100mm or 150mm in length. Please contact us for technical assistance.

Hypersil GOLD PFP (perfluorophenyl) columns build on the performance of the Hypersil GOLD columns by providing excellent peak shapes while also offering alternative selectivity in reversed phase chromatography compared to alkyl chain phases. The Hypersil GOLD manufacturing process provides improvements in speed of analysis, peak shape and sensitivity over other fluorinated phases. This will be of particular interest when trying to analyze compounds which are difficult to retain or which have complex halogenated functionality.
Unique Selectivity
Hypersil GOLD PFP columns help to extend the range of selectivities that are provided in reversed phase chromatography. This phase will increase retention and resolution, and is particularly suited to separation of halogenated species as well as compounds containing hydroxyl, carboxyl, nitro or other polar groups. The additional mechanisms of the PFP phase, such as steric selectivity of the analyte species and dipole interactions, offer alternative selectivity. Perfluorinated phases like Hypersil GOLD PFP are frequently used in the analysis of complex taxane samples.

PRISM RP Columns
PRISM RPN columns are HPLC columns that are ideal for 0 to 100% organic fast gradients.
- Polar embedded C12 phase for unique selectivity

For alternative selectivity to traditional alkyl phases, we recommend you try the polar embedded phases of PRISM columns. The stationary phase contains urea groups embedded within the C12 chain, which extra interactions with samples to extend the capabilities of the phase.
PRISM RP is an endcapped stationary phase, based on a highly base deactivated 100Å pore size silica. The pore size combined with the C12 phase gives the columns moderate hydrophobic retention for neutral samples, but compared to the conventional phases (C18 or C8) it offers a completely new selectivity.
A polar functional group between alkyl ligands and the silica surface allows fast analysis without compromising the resolving power. PRISM RP is completely stable under 100% aqueous conditions, so it is ideal for 0 to 100% organic fast gradients.
