Tosoh Bioscience offers a broad line of high efficiency columns for analysis and isolation of biomolecules by ion exchange chromatography. For Anion as well as Cation Exchange Chromatography, the product line contains methacrylate-based and silica-based columns for the analysis of proteins, peptides, DNA- and RNA- derived oligonucleotides and other nucleic acid fragments.

Polystyrene-based ion exchange columns are available for analyzing low MW sugars, amino acids, nucleic acid bases, and small drug candidates.

Hydrophilic polymer-based, exchange columns packed with non-porous resin particles are available for the separation of proteins, protein aggregates, charge isomers of monoclonal antibodies, PEGylated proteins, DNA fragments, nucleic acids, oligo DNA, and siRNA.

Specialty columns are available for organic acids by ion-exclusion (OApak-A and SCX) and mono- and disaccharides (Sugar AXI and Sugar AXG).

Common Characteristics of TSK-GEL Anion and Cation Exchangers

The methacrylate base resin, G5000PW (5PW), is a spherical 10μm particle with approximately 1000Å pores. TSK-GEL 5PW is derivatized with diethylaminoethyl (DEAE), sulfopropyl (SP) or carboxymethyl (CM) functionalities to provide a weak anion, a strong cation, and a weak cation exchanger, respectively. The polyamine chemistry employed in TSKgel SuperQ-5PW results in a high capacity strong anion exchanger with a smaller effective pore size than TSKgel DEAE-5PW.

Particles in TSKgel BioAssist Q columns contain very large pores (~4000Å) that are functionalized with polyamine groups to form a network structure. The capacity of BioAssist Q columns is high over a wide molecular weight range. TSK-GEL BioAssist S columns are packed with particles possessing 1300Å pores functionalized with sulfopropyl groups.

Methacrylate is also the backbone of non-porous resin (NPR) columns such as DEAE-NPR, DNA-NPR and SP-NPR, which are packed with 2.5μm particles. High column efficiency coupled with low sample capacity restricts the application of these columns to fast analysis and micro-scale preparative isolation. Due to the absence of large pores, protein recovery is generally very high on NPR columns.

TSK-GEL STAT Series non-porous resin columns contain large particles (5, 7 and 10μm). This relatively large particle size, combined with an innovative bonding chemistry, results in columns that enable fast equilibration and analysis of complex biomolecular samples.

Anion Exchange Columns

Anion exchange chromatography is practiced with either a strong or a weak anion exchange column, containing a quarternary ammonium ion, or with a weak anion exchanger, having either a tertiary or secondairy amine functional group, such as DEAE (diethylaminoethyl). A counter ion, often Cl- maintains electroneutrality.

In ion exchange chromatography the pH of the mobile phase buffer must be between the pI or pKa of the charged molecule and the pKa of the charged groups on the solid support. For instance, in anion exchange chromatography a molecule with a pI of 6.8 is run in a mobile phase buffer at pH 8.0 with the solid support pKa at 10.3.

  • Binding capacity for small to medium size proteins on TSKgel DEAE-3SW is roughly double that of the 5PW packings due to the smaller pore size and larger surface area.
  • TSKgel DEAE-5PW and DEAE-2SW columns are available in 2mm ID format
  • Specialty columns for analysis of mono- and disaccharides, sugar alcohols, and organic acids.

  • BioAssist columns are packed in PEEK hardware, while other columns are available in glass and stainless steel.
  • Very efficient chromatography can be achieved with the TSK-GEL STAT columns due to the novel bonding chemistry and the absence of micro-pores.

TSKgel BioAssist Q

TSKgel BioAssist Q columns are packed with spherical, polymer beads with approx. 4000Å pores. They are surface modified with a strong anion exchanger. The very large pores allow large biomolecules to freely enter and exit the pores resulting in higher binding capacity and better recovery of activity than other competivie materials. The standard column is 4.6mm ID x 5cm, while a 10mm ID x 10cm semi-preparative column is available for scale-up. Both columns are made of PEEK to reduce protein adsorption.

Cation Exchange Columns

Cation exchange chromatography is practiced with either a strong or a weak cation exchange column, containing a sulfonium ion, or with a weak cation exchanger, having usually a carboxymethyl (CM) functional group. A counter ion, often Na+ maintains electroneutrality.

In ion exchange chromatography the pH of the mobile phase buffer must be between the pI or pKa of the charged molecule and the pKa of the charged groups on the solid support. For example, a molecule with a pI of 8.2 is run in a mobile phase buffer at pH 6.0 with the solid support pKa at 1.2 in cation exchange chromatography.

Highlights

  • Pore structure and bonding chemistry of TSKgel BioAssist S provides high capacity for medium to large molecular weight proteins.
  • BioAssist columns are packed in 4.6mmID or 10mmID PEEK hardware.  Other columns are available in glass and stainless steel for anlytical, semi-preparative and preparative applications.
  • Binding capacity  for small to medium size proteins on TSKgel DEAE-3SW is approximately double that of the 5PW packings due to the smaller pore size and larger surface area.

TSKgel BioAssist S

TSKgel BioAssist S columns are packed with porous hydrophilic polymer beads which are surface modified with a strong cation exchanger. The resin's large pores are very accessible even for high molecular weight proteins. This leads to higher chromatographic efficiency and binding capacity for purification.

The application on this page shows the analysis of bromelain, a proteolytic enzyme that is used as a nutritional supplement. Bromelain is a basic glycoprotein with a MW of 33kDa and pI of 9.55.

 Applications


 The analysis profiles for five antibodies on a TSKgel CM-STAT column were compared with the profiles obtained on a competitive non-porous cation exchange column. Similar or higher resolution profiles were obtained on the TSKgel CM-STAT column in approximately half the time.
