TSK-GEL SW-type columns for Quality Control of Proteins

TSK-GEL SW-type columns are silica-based. A hydrophilic diol-type bonded phase shields the silica surface from interacting with protein samples. Three different pore sizes are available.

  • 125Å pore size for analysis of small proteins and peptides
  • 250Å pore size for most protein samples
  • 450Å pore size for very large proteins and nucleic acids

Compared to competitive HPLC columns, SW-type columns contain more pore volume per unit column volume, which results in higher MW selectivity or better resolution.

Particle sizes for the three SW-type column lines are:

  • SuperSW: 4 micron
  • SWxl: 5-8 micron
  • SW: 10-13 micron

Highlights

  • TSK-GEL SW-type packings are comprised of rigid spherical silica gel particles chemically derivatized with diol-containing ligands.
  • Low adsorption and well-defined pore size distribution, which are requirements for high performance Gel Filtration of proteins.
  • Three pore sizes are available in SW or SWxl product lines, two in the SuperSW product line 
  • TSK-GEL SuperSW columns feature 4 micron particles for the highest resolution. The narrow 4.6mm column ID provides the highest sensitivity when limited in sample mass.
  • Semi-preparative (21.5mm ID) and preparative (55mm, 108mm ID) stainless steel columns are available for precise scale-up to commercial production.

TSK-GEL SuperSW

TSK-GEL SuperSW columns, introduced in 1997, represent the third generation of high performance Gel Filtration columns that have become synonymous with analyzing protein molecular weights in the emerging field of biotechnology.

Particle Characteristics
TSK-GEL SuperSW-type columns contain smaller particles than SWxl-type columns; 4 micron versus 5 micron. In addition, the column internal diameter has been reduced from 7.8mm ID to 4.6mm ID to provide higher sensitivity in sample-limited cases and to cut down on solvent use.

HPLC System Requirements
It is important to employ an HPLC system that is optimized with regards to extra-column bandbroadening to take full advantage of the high column efficiency that can be obtained on SuperSW columns. See Separation Report 95 for recommendations.

Calibration Curves
The figure to the right shows the calibration curves for globular proteins for the two TSK-GEL SuperSW-type columns in comparison with those for the corresponding SWxl columns. The following table summarizes the molecular weight ranges for SuperSW2000 and SuperSW3000 columns for globular proteins, polyethylene oxides and dextrans that can, at least partially, access the pores. For comparative purposes, similar data is provided as well for TSK-GEL SW and SWxl-type columns.

TSKgel SuperSW2000

To fully benefit from the high efficiency of 4μm columns, and because the column diameter of TSK-GEL SuperSW columns is 4.6mm, it is important to minimize the amount of dead volume in the HPLC system. Consult the Separation Report for details (provided below).

TSK-GEL SWxl-type columns contain smaller particles than SW-type columns; 5 and 8μm versus 10 and 13μm. As the SW-type columns, TSK-GEL SWxl columns feature highly porous silica particles, the surface of which has been shielded from interacting with proteins by derivatization with ligands containing diol functional groups. TSK-GEL SWxl-type columns stand out from other silica- or polymer-based high performance size exclusion columns by virtue of their large pore volumes. These columns are commonly used in the quality control of monoclonal antibodies and other biopharma products.

The Figure below shows the calibration curves for globular proteins, polyethylene oxides and dextrans for each of the three TSK-GEL SWxl-type columns. TSKgel G2000SW

As the first generation of silica-based SEC columns, TSKgel G2000SW provides excellent separation of peptides and proteins with molecular weights up to 100,000 Da.

TSK-GEL G2000SW columns start at 10μm for analytical columns and also scale to 13 and 20μm in columns up to 55mm ID for preparative purifications.

TSK-GEL G3000SW

As the first generation of silica-based GFC columns, TSK-GEL G3000SW columns provide excellent separation of proteins and other biomolecules with molecular weights up to 500,000 Da.

The TSKgel G3000SW columns start at 10μm for analytical columns and also scale to 13 and 20μm in columns up to 55mm ID for preparative purifications.

TSK-GEL G4000SW

As the first generation of silicia based SEC columns, the TSK-GEL G4000SW columns provide excellent separation of proteins and other biomolecules.

TSK-GEL G4000SW columns are available in 13μm particle size in Glass and Stainless Steel analytical columns. Particle sizes of 13 and 17μm are used to pack 30cm and 60cm x 21.5mm ID columns, respectively, for semi-preparative purifications.
